龟板浸膏甲酯化GC-MS干细胞活性指纹图谱的研究与应用

摘  要: [目的]为了确定运用生物活性指纹图谱预测中药的干细胞活性的可靠性而进行了该试验。[方法]试验中,依次用石油醚、乙醚、二氯甲烷提取龟板浸膏,得到溶剂提取物,将一部分溶剂提取物甲酯化,并用MTT比色法及流式细胞仪研究了甲酯化产物的干细胞活性;采用GC-MS技术研究每一个甲酯化产物,根据总离子流色谱图的相同保留时间和相应峰面积,获得龟板浸膏甲酯化GC-MS生物活性(干细胞活性)指纹图谱。结果表明:甲酯化产物均能促进干细胞的体外增殖,用龟板浸膏甲酯化GC-MS生物活性指纹图谱来预测中药"四物汤"的干细胞活性,平均预测准确率达87.5%。[结论]因此,用这种方法来预测未知中药样品的干细胞活性,有一定的可靠性。 The experiment was done to confirm the reliability of applying biological activity fingerprint to predict the stem cell activity of traditional Chinese medicine. In the experiment, the solvent extracts were obtained through extracting with petroleum ether, aether and methylene chloride in tuna from tortoise shell extract, then some solvent extracts were methyl-esterified and the stem cell activities of methyl-esterification products were studied by MTT coloriraetry and flow cytometry. Every methyl-esterification product was studied by GC-MS technology and the CG-MS biological activity (stem cell activity) fingerprint of metyl-esterified tortoise shell extract was obtained on the basis of the same retention time and the corresponding peak area of total ion chromatogram. The results showed that all the methyl-esterification products could promote the proliferation of stem cell in vitro and the average prediction accuracy of predicting the stem cell activity of traditional Chinese medicine Siwu decoction by the CG-MS biological activity fingerprint of metyl-esterified tortoise shell extract was up to 87.5%. So, predicting the stem cell activity of unknown traditional Chinese medicine sample by this method had some reliability.
