摘 要: 在结合国内外研究成果的基础上,采用超声强化样品前处理同时对液相色谱的分离条件进行优化。超声提取条件:温度30℃,样品:溶剂=1:15(质量与体积或体积与体积之比)避光提取(30min+15min)2次;色谱条件为色谱柱:(Inertsil ODS-3,C_(18)反向柱,4.6mm×250mm,5μm);流动相:V(甲醇):V(三氯甲烷)= 65:35;紫外检测器波长450nm;流速1.2mL/min;柱温30℃。结果表明:该方法具有提取率高、操作时间短、溶剂用量少,灵敏度高、准确度好和可靠性强等优点。 This text combines the foundation of the domestic and international research result. Besides adopting the Ultrasonic Extraction to enhance the sample handle ,the experiment also optimize the condition which β-carotene can be separated from other substance by HPLC. The ultrasonic extraction condition: 30℃, sample: solvent =1 : 15(m : v or v : v) , (30 +15 min) two times. The HPLC condition: Inertsil ODS-3 ,C18 (416 mm×250 mm,5 μm),UV detector set at 450nm, mobile phase .V (CH3OH) : V (CHCl3) =65 : 35 and flow at 1.2 mL/min. The experiment showed that this method is effectiveness, short time, less solvent, sensitive, accurate and reliable.
