摘 要: 离子通道的开闭反映了蛋白质构象变化的动力学过程。单通道膜片钳记录是有关通道蛋白动力学性质的主要信息来源。对开闭时间长短的分析可以揭示其门控机制。当今,齐次马尔可夫过程常常用来分析膜片钳实验的数据,但未经检验齐次性,不能拿来即用。我们给出了一个新的模型,称之为非齐次马尔可夫模型,通过它,可以把时间和速度常数roc与rco联系起来,把roc写作roc(t),把rco写作rco(t),而不再当成常量。我们给出了一种估计方法,并对模拟数据进行了参数估计,统计结果表明本方法是可行的。 Openings and closings of channel reflect the dynamics of protein conformations.Single-channel patch clamp recordings are principle sources of information about dynamical properties of channel proteins.Analisis of the durations of the open and closed intervals can give insight into the gatng mechanism. Now,homogeneous Markov process is usually used to analise the data from patch clamp experiment,but we can not use it at once without examining the hypothesis about homogeneous.We give a new model called Non-homogeneous Markov model,through which we can link the time and the rates,r oc and r co ,instead of contants,we write r oc as r oc (t),r co as r co (t).We provide a method about evaluating,and evaluated the parameters,the data was from simulatons.Statistics shows that our method is feasible.
