摘 要: 采用HPLC-UV与反相C18柱,以50 mmol/L的KH2PO4(pH 5.5)-10%的甲醇(V/V)作为洗脱液,同步快速检测了21位健康人与90位病人胃粘膜活检组织中的脱氧鸟苷(deoxyguanosine,dG)和8-羟基脱氧鸟苷(8-Hydroxydeox-yguanosine,8-OHdG)的量。方法线性范围分别为3.0~400μg/mL、0.2~10μg/mL,检出限可分别达到1.0、0.1μg/mL(S/N=3)。实验发现病人胃粘膜活检组织中的8-OHdG量远远高于健康人,此结果在临床早期诊断相关疾病方面具有很高的应用价值。 8-Hydroxydeoxyguanosine (8-OHdG) has been widely used as a biomarker of oxidative DNA damage in livings. However, the levels of 8-OHdG aren't available on other exposure such as human gastric mucosal biopsy. In this paper, we simultaneously investigated the levels of 8-OHdG and dG in DNA from human gastric mucosal biopsy with HPLC-UV. The analysis was performed on a RP-18 analytical column using potassium phosphate buffer (50 mmol/L, pH 5.5)-methanol (90/10, V/ V) as eluent. The calibration graph for 8-OHdG was linear in the concentration range of 0.2 - 10μg/mL, and the limit of detection was 0.1μg/mL(signal to noise ratio S/N = 3). The concentration of 8-OHdG in 21 healthy persons and 90 patients' gastric mucosal biopsy was determined. It was found that levels of 8-OHdG in patients was significantly higher than those in healthy persons (p 〈 0. 001 ), the results demonstrated that 8-OHdG may be a good marker for diagnosis of some illness.
