摘 要: 研究超声波提取柿叶中总黄酮的工艺。采用超声波提取法对柿叶中总黄酮进行提取,通过单因素及正交实验,确定最佳工艺。随着乙醇浓度的增大,柿叶总黄酮的提取率增大,当乙醇浓度超过60%后提取率下降。总黄酮提取率随料液比的增大而迅速增大,当料液比达1∶20时,增加幅度明显减缓;总黄酮提取率随超声波提取时间的增加而呈上升趋势,但趋势不明显。影响柿叶总黄酮提取率因素的顺序为料液比>提取时间>乙醇浓度,最佳提取工艺是乙醇浓度60%,超声波提取时间45 min,料液比1∶25,总黄酮得率为3.54%。与常规回流法相比,超声波提取法柿叶中总黄酮的得率提高77%,并缩短了提取时间,降低了提取溶剂的用量。超声波提取法具有提取效率高、成本低的明显优势。 The purpose was to study the technology of extracting total flavones from Persimmon leaves by ultrasonic wave.Total flavones were extracted from Persimmon leaves by ultrasonic wave.The optimum technology was ascertained through single factor and orthogonal experiments.The extraction rate of total flavones from Persimmon leaves was increased along with the increment of ethanol concentration.The extraction rate was reduced after the ethanol concentration exceeded 60%.The extraction rate of total flavones was rapidly increased along with the increment of the ratio of material to liquid.The increasing amplitude was obviously slowed when the ratio of material to liquid reached 1∶20.The extraction rate of total flavones was increased along with the increment of ultrasonic extracting time,but the trend was not obvious.The order of factors that affected the extraction rate of total flavones from Persimmon leaves was the ratio of material to liquid 〉 extraction time 〉 ethanol concentration.The optimum extraction technology was as follows: ethanol concentration of 60%,ultrasonic extracting time of 45 min,ratio of material to liquid of 1∶25,under which the extraction rate yield of total flavones was 3.54%.Compared with conventional refluxing method,the yield of total flavones from Persimmon leaves by ultrasonic wave was enhanced by 77%.And this method shortened the extraction time and reduced the dosage of extraction solvent.Ultrasonic wave extraction method had obvious advantages of high extraction efficiency and low cost.
