avUCP基因与日粮能量水平对鸡骨骼肌ATP含量的影响
Effects of Avian UCP Gene and Dietary Energy Levels on Skeletal Muscle ATP Content of Chickens

摘  要: 探讨鸡在不同日粮能量水平下,鸡解耦联蛋白(avian uncoupling protein,avUCP)基因对骨骼肌组织中ATP含量的影响。试验采用2×3×3三因子完全随机试验设计,即avUCP转基因鸡与非avUCP转基因鸡、3种日粮(高能日粮、高脂日粮和标准日粮)和日粮饲喂时间(20、40和60 d),共18个处理组。试验期60 d,试验结束时,屠宰试鸡取骨胳肌组织,生物发光法测定骨骼肌ATP含量,荧光定量PCR检测avUCP mRNA的表达水平。结果表明,avUCP基因、日粮营养结构和日粮饲喂时间均显著影响鸡骨骼肌的ATP含量(P<0.01),且ATP含量与avUCP mRNA的表达水平显著负相关(P<0.05),相关系数为0.45,avUCP基因和日粮营养结构、日粮营养结构和日粮饲喂时间之间均存在显著的互作效应(P<0.05)。avUCP转基因鸡骨骼肌ATP含量明显高于非avUCP转基因鸡,其中高能日粮组间差异极显著(P<0.01),高脂日粮组间和标准日粮组间差异显著(P<0.05)。3种日粮处理显著影响骨骼肌ATP含量(P<0.01),其中avUCP转基因组的差异比非转基因组大。高能日粮组和高脂日粮组饲喂前期骨骼肌ATP含量逐渐增加,40 d时达到最高,其后呈降低趋势(P<0.05)。通过日粮营养和转基因技术调节avUCP mRNA的表达可以促进骨骼肌ATP的储备,在细胞和分子水平上节省鸡的能量消耗。 The aim of this study was to probe into the effects of avian UCP gene on skeletal muscle ATP content for chickens in different energy diets. 2 × 3 × 3 three-factor variance analysis experiment was designed, including avian UCP transgenic chickens and avian UCP non-transgenic chickens, three diets (high energy diet, high fat diet, standard diet) and three dietary feeding periods( 20 days, 40 days, and 60 days), consisting of 18 treatments. The experiment lasted for 60 days, when the experiment ended, all chickens were killed to separate skeletal muscle for assaying ATP content by Bioluminescence. Furthermore, avian UCP mRNA expression was determined in skeletal muscle by reverse transcription-polymerase chain reaction. The results showed that the ATP content in the skeletal muscle correlated with avian UCP mRNA expression negatively (P〈0.05), the coefficient was 0.45, the effect of interference was significant between avUCP gene and nutrition structure, nutrition structure and raise time (P〈0.05). The skeletal muscle ATP contents of avian UCP transgenic chickens were higher than those of non-transgenic chickens among all dietary groups, the diversity of high energy dietary group was significant (P〈0.01), that of high fat dietary and standard dietary group was significant (P〈0.05). The treatments among three diets were significantly different, and the ATP contents of skeletal muscle in high energy groups were higher than those of the other two dietary groups (P〈0.01), furthermore, the differences of transgenic groups were more significant than those of non-transgenic groups. The skeletal muscle ATP contents of 40-day feeding period were higher than those of 20-day and 60-day feeding period in high energy and high fat dietary groups (P〈0.05), so it might approach the highest status in the 40th day of treatment.
