摘 要: 对转基因植株T0代种子进行发芽试验以及对T1代植株叶片进行浸泡试验,以研究转基因植株后代的遗传及表达情况。结果表明,转基因植株T0代种子在含50 mg·L-1潮霉素的培养基上正常发芽生长,而对照虽能发芽,但最终枯黄死亡;T1代植株叶片在100 mg·L-1潮霉素溶液中浸泡3 d后,部分叶片变黄,部分叶片仍为绿色,统计分析表明,转基因植株外源片段为单拷贝整合;对潮霉素浸泡后的植株的PCR分析表明,潮霉素浸泡对后代的遗传分析是行之有效的。 For genetic analysis of transformants, the To generation seeds of the transgenic plants were germinated and the leaves of T1 generation plants were soaked. The result indicated: the seeds of the transgenic plants could germinate and grew very well on the MS medium with 50mg·L^-1 hygromycinB. The seeds of the non-transgenic plants also could germinate, but its seedlings wilted and died finally; the segregation analysis of Hyg-resistant in T1 progeny showed the foreign fragments was one locus; soaking the leaves with hygromycin B was proved to be an useful method for genetic analysis of progeny by the correspondent PCR analysis.
