摘 要: 为了研究异丙醇对猪胰脂肪酶催化橄榄油水解反应的作用机理,在体系中加入不同体积分数的异丙醇,测定了脂肪酶的活性、动力学和光谱变化。活性测定结果表明,低体积分数的异丙醇对该酶有一定的激活作用,体积分数为20%的异丙醇使酶活性提高了229%。光谱分析结果显示,经异丙醇处理后猪胰脂肪酶的紫外吸收光谱和荧光发射光谱发生了变化,紫外差示光谱则出现了明显的负峰。动力学研究结果表明,异丙醇处理后,猪胰脂肪酶的vmax基本不变,而Km值则明显减小。 In order to study the mechanism of isopropanol on porcine pancreas lipase(PPL)activity,different volume fractions of isopropanol were added into the reaction system of olive-oil hydrolysis to test PPL activity, kinetics and spectrum. It was indicated that PPL could be activated by isopropanol at low volume fraction. The hydrolysis activity of PPL was increased by 229% when the system Contained 20% isopropanol. After PPL was treated by isopropanol, both the ultraviolet absorption spectrum and fluorescent emission spectrum were changed,and the differential ultraviolet spectrum showed negative peaks. When PPL was treated with isopropanol,the vmax remained unchanged while Km value declined.
