摘 要: 利用RT-PCR法和PCR法克隆到橄榄星室木虱乙酰胆碱酯酶基因(Ace)的2个片段,其中cDNA片段长66bp,翻译获得的氨基酸1~11位与家蝇等13种昆虫相应的序列相同或仅相差1个氨基酸,该序列在GenBank数据库中的接受号为DQ417582;基因组DNA片段长499bp,2个外显子连在一起与意大利蜜蜂等13种昆虫相应基因的氨基酸同源性均高于88%,该序列在GenBank数据库中的接受号为DQ425028。此结果为进一步研究橄榄星室木虱对杀虫剂抗性的分子机理奠定了基础。 RT-PCR and PCR were used to clone the acetylcholinesterase gene (Ace)of Pseudophacopteron canarium. Two fragments of the gene were obtained. One of them, a cDNA fragment with the length of 66 bp, was deposited in GenBank with the accession number of DQ417582.Among 1 to 11 of the amino acid residues translated from DQ417582, there was no or only one amino acid difference between the CDNA fragment of P. canarium and corresponding gene of other 13 insect species. The other fragment of the Ace, a genomic DNA with the length of 499 bp, was deposited in GenBank with the accession number of DQ425028. The similarity of the amino acid residues translated from two exons combined together in DQ425028 was more than 88% compared with corresponding gene of other 13 insect species. This research is helpful for further studies on molecular mechanism of the resistance of P. canarium to insecticides.
