摘 要: 目的研制丙型肝炎病毒(HCV)基因分型寡核苷酸(Oligo)芯片并进行杂交验证。方法分别对BLAST检索所得的HCV 4个亚型型特异序列逐一进行分析,设计长度均一的60mer Oligo探针,用芯片点样仪将设计好的探针打印到玻片上制备成基因芯片。结果杂交结果显示,Oligo芯片检测HCV各基因亚型的效果较佳。结论制备的长链Oligo分型芯片检测敏感性、特异性均为满意,为下一步集合更多种肝炎病毒的联合检测与分型打下基础。 Objective A model microarray assay for the detection of hepatitis C virus (HCV) genotypes were developed. Methods 60-mer Oligo probes were designed according to the conserved region of different genotypes of HCV after BLAST. The DNA microarray was prepared by spotting Oligo probes on the surface of amido-modified glass slide using the robotics system. The detection of microarray was validated by the hybridization and the sequence analysis. Results The diagnostic validity of the microarray method was evaluated after the washing and scanning process. The results of hybridization and sequence data analysis showed the specificity and sensitivity in the detection of HCV genotypes were satisfied. Conclusion Using 60-mer Oligo probes to construct gene chips for clinical diagnosis of HCV genotypes resulted in a rapid, simple and effective method. That the Oligo microarray assay may be widely used in the clinical diagnosis of hepatitis virus and genotypes.
