摘 要: 为探讨电压依赖性钾通道亚基Kv2.1、Kv3.1b在神经毒素鱼藤酮及神经肽PACAP27作用下可能的表达变化,本研究采用定量逆转录多聚酶链反应(RT-PCR)和蛋白免疫印迹分析(Westernblot)检测了PC12细胞Kv2.1、Kv3.1bmRNA及蛋白水平的表达。结果显示:(1)鱼藤酮可使Kv2.1mRNA的表达增高(P<0.05),但同时却又使Kv2.1蛋白水平的表达降低(P<0.01),PACAP不能逆转其作用(P>0.05);(2)鱼藤酮及PACAP27对Kv3.1bmRNA的表达无明显影响(P>0.05),但鱼藤酮却可使Kv3.1b蛋白水平的表达降低(P<0.05),PACAP27可以拮抗鱼藤酮导致的蛋白降低(P<0.01)。以上结果提示神经肽PACAP27及鱼藤酮对于Kv2.1、Kv3.1b表达的影响在mRNA及蛋白水平存在着差异,PACAP的保护作用可能与其在蛋白水平增加Kv3.1b的表达有关。 To observe the selective alterations of Kv 2.1 and Kv 3.1 b expression in PC12 cells induced by neurotoxin-rotenone and neumpepdde-PACAP, quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western blot were performed to detect alterations of Kv 2.1 and Kv 3. lb expression in PC12 cells at mRNA and protein level. Our data demonstrated : ( 1 ) rotenone can increased significantly expression of Kv 2.1 at level of mRNA ( P 〈 0.05 ) , however inhibited expression of Kv 2.1 at level of protein ( P 〈 0.01 ). By contrast, PACAP had no effects on expression of Kv 2.1 (P 〉0.05) ; (2)rotenone significantly inhibited protein expression of Kv 3. 1 b ( P 〈 0.01 ) , but no effects on expression of Kv 3.1 b at level of mRNA ( P 〉 0.05 ). Although PACAP had no effects on Kv 3.1 b at level of mRNA, but significantly improved the protein expression of Kv 3.1 b against rotenone-induced inhibitory effects ( P 〈 0.01 ). Our study indicates that rotenone and PACAP27 have different effects on expression of Kv 2.1 and Kv 3. lb in mRNA and protein level, and the protection of PACAP might concerned with enhancing the protein expression of Kv 3.1 b.
