摘 要: 综述近年来膜蛋白结构预测的研究进展,分类介绍了α螺旋和β桶状跨膜蛋白结构预测方法之基本原理和应用。作为重要的基因产物之一,跨膜蛋白在生物体中发挥各种重要功能,而膜蛋白偶联受体是大多数药物的作用靶点。基于结构的药物设计是目前药物研发的主要方法,可膜蛋白的三维结构很难采用X-光晶体衍射和核磁共振技术测定,因此应用生物信息学方法预测膜蛋白结构成为当今研究热点。 The advances in membrane protein structure prediction in recent years were reviewed. The basic principle and application of the structure prediction methods of α-helix and β-barrel membrane proteins were respectively introduced. As one of the most important genetic products, membrane proteins play multiple important roles in organisms. Furthermore, the membrane-protein-coupled receptors are the targets of most medicines. At present, structure-based drug design is the main method of drug development. However, the high-resolution experimental structures of membrane proteins remain exceptional to be difficult to be determined by X-ray disffraction and NMR. As a result, using bioinformatics to predict the structures of membrane proteins is intensively focused.
