摘 要: 利用随机扩增多态DNA(RAPD)技术分析了斜带石斑鱼Epinephelus coioides繁育亲本以及2001、2004年孵育的2批子代群体的遗传多样性。17个引物共扩增出104个位点。应用生物软件POPGENE进行统计分析,3个取样群体的多态位点百分率分别为32.69%(繁育亲本)、30.77%(2001年子代)和24.04%(2004年子代);基因多样性和香侬信息指数均为繁育亲本群体最高(0.110 8和0.167 0)、2004年子代群体最低(0.066 6和0.103 8)。通过t检验检测群体间遗传多样性的差异显著程度并根据Nei公式计算遗传相似度和遗传距离分析群体间的遗传分化。结果表明,RAPD技术具有较高的灵敏性和多态性,是分析评估斜带石斑鱼遗传多样性较为适宜的分子标记之一;人工养殖不仅使斜带石斑鱼遗传多样性逐年下降,而且目前已经降低到显著的程度,近交和遗传漂变可能是导致遗传多样性下降的2个重要因素。最后对保持斜带石斑鱼遗传多样性的养殖策略进行了探讨。 Random amplified polymorphic DNA (RAPD) technique was employed to analyze the genetic diversity of the brood stock of Epinephelus coioides and their two filial generation stocks hatched in 2001 and 2004. Total 104 loci were amplified using 17 random primers. The percentages of polymorphic loci of those three stocks were calculated by software POPGENE as 32.69%, 30.77%, and 24.04%, respectively. The gene diversity and Shannon's information index of the brood stock were the highest (0.110 8 and 0.167 0), and the filial generation stock hatched in 2004 had the lowest values (0. 066 6 and 0. 103 8). In addition, t-test was used to test the difference significance of genetic diversity among the stocks, and the genetic identity and genetic distance were calculated according to Nei's formula to analyze the stock differentiation. The results showed that RAPD technique, with its high sensitivity and polymorphism, was suitable for the study on genetic diversity of E. cofoide; a gradual decline of genetic diversity of E. coioides had been up to a remarkable level in aquaculture practices; and inbreeding and genetic drift were two likely important factors causing the decline of genetic diversity. Furthermore, some suitable strategies were discussed for genetic diversity conservation of E. coioides.
