摘 要: 用液氮熏蒸法在氟板上制作冻精颗粒,以解冻后的精子活率、活力和质膜完整性为判定指标,比较4种冷冻稀释液及不同冷冻-解冻程序对五指山小型猪精液冷冻的效果。结果表明:①Ⅳ号冷冻稀释液冷冻解冻后精子的活率(0.610±0.036)、活力(0.427±0.025)和质膜完整性(0.503±0.015)均显著高于Ⅰ、Ⅱ和Ⅲ号冷冻稀释液(P<0.05)。②实验中精液在4℃冰箱中平衡降温2 h的精液精子活力、质膜完整性均好于在17℃平衡3 h再放入4℃冰箱中平衡2 h的解冻效果,而且精子活率差异显著(P<0.05)。③湿解法的效果优于干解法。 Frozen boar semen pellets were made on the fluorin plant. To select out a best freezing extender from several extenders and freezing-thawing protocols, sperm viability, motility and plasma membrane integrity were studied in this experiment. The results showed that(1)the number Ⅳ extender was the best among the extenders tested and its sperm motility (0.427 ± 0.025), viability (0.610 ± 0.036) and plasma membrane integrity (0.503 ± 0.015) after-thawing were all significantly higher than those obtained with number Ⅰ, Ⅱ and Ⅲ extenders(P 〈 0.05 ); (2) The sperm motility and plasma membrane integrity of semen cooling for 2 h at 4℃ after-thawing were both better than those cooling for 3 h at 17℃ and then transferring in 4℃ refrigerator for 2 h. And the sperm viability had significant differences(P 〈 0.05 ); (3)The effects of thawing in solution were better than those out of dried thawing.
