摘 要: 目的:优化乙肝传感器中抗体的固定条件以制得免疫传感器。方法:以金电极为基底,自组装半胱胺,通过戊二醛交联法将抗体蛋白固定在电极上,制成乙肝修饰电极。以Fe(CN)63-/Fe(CN)64-为电活性指示剂,利用循环伏安法考察了该修饰电极在不同阶段的电化学性能。结果:结果显示优化抗体固定条件后的传感器能对乙肝HBsAg阴阳性抗原进行区分检测。结论:用半胱胺/戊二醛方法固定乙肝抗体制得免疫传感器能与相应的抗原发生免疫反应,具有实际应用的前景。 Objective:To optimize the immobility condition of HBV antibody to make hepatitis B biosensor. Methods:The HBV antibody was immobilized on the gold surface with self- assembled. The electrochemical characteristics of the modified electrode at different stages of modification was studied by cyclic voltammetry (CV) in the Fe(CN)6^3-/Fe(CN)6^4- solution. Results: The negative and positive HBsAg antigen could be devised. Conclusion :The hepatitis B biosensor made by cystamine/glutaraldehyde assembled can immunoreact with the corresponding antigens. And the methode to make biosensor has its practicality.
