摘 要: 通过实验测定,证明生物反应器中细胞死亡速率与气体鼓泡速率成正比而与反应器体积成反比。实验发现气泡大小对细胞死亡速率具有两种作用,一种作用在于影响气泡表面积生成速率;另一种作用则在于影响细胞在气泡表面的吸附程度,其最佳直径为5mm左右。血清和Pluronic P68能显著降低细胞死亡速率,当Pluronic F68浓度达到0.1%时,k_d趋于零。所有这些实验结果均与前文提出的生物反应器设计模型具有很好的一致性。 The experimental results obtained from this work demonstrated that the cell death rate in sparged bioreactors is proportional to the gas sparging rate and inversely proportional to the bioreactor volume. It was found that the bubble size has two effects on cell death rate. The one is to affect the bubble surface area production rate and the other is to influence the adsorptivity of the cells on bubble surface. The optimum bubble diameter at which the cell death rate reaches minimum is about 5mm. The effect of media formulations on cell damage was also investigated. Both the serum and Pluronic F68 were found to reduce the cell death rate significantly. And as the concentration of Pluronic F68 increases to 0. 1% , the value of kd drops to zero. All the experimental data are in excellent agreement with the bioreactor design model developed in the previous paper.
