利用甘蔗糖蜜发酵生产酵母单细胞蛋白的菌种选育

摘  要: 以甘蔗糖蜜作为发酵原料,经过摇瓶发酵培养,筛选得到一株总蛋白得率较高的啤酒酵母(Saccharomycescerevisiae)S9。以S9为出发菌株进行60Co诱变选育实验,筛选到一株高生物量高蛋白含量的S906突变菌株,在相同的发酵条件下,生物量为11.25g/L,蛋白质质量分数为45.96%,总蛋白质得率达5.17g/L,比诱变前增加了39.0%。S906传代10次,其生物量、细胞粗蛋白含量无大的变化,表明该菌株遗传性状稳定,经过改良可作为工业化生产单细胞蛋白菌株。 Using sugar cane molasses as fermentation substrate, a Saccharomyees cerevisiae strain S9 was obtained after fermenting and gradual screening, and then treated with ^60Co radiation to get a positive mutation yeast strain S906 that contained more biomass and protein. Under the same fermentation conditions, the biomass was up to 11.25 mg/L, protein content of the yeast celt was up to 45. 96% and total protein content amounted to 5.17 mg/L, which was 139% higher than that before mutation. Strain S906 was transferred 10 times, but its biomass and crude protein content in cells were changed not greatly. The result showed that this strain had stable genetic characteristics and could be used for industrial production of single cell protein after improvement.
