摘 要: 运用农杆菌转化法将叶片衰老抑制基因PSAG12-ipt转入南方推广的优质籼型不育系中A、恢复系明恢63、南胜10号,所获得的潮霉素抗性植株通过GUS组织化学分析、PCR检测、Southern blot分子检测证实,PSAG12-ipt已经转入水稻基因组中。对转基因植株进行了延衰性考察,结果表明,与对照组相比,部分转基因水稻植株明显表现出叶片推迟衰老,提高了单株有效穗数、千粒重。对转基因R1代植株进行了PCR和PCR Southern分析,表明外源基因在转基因植株后代中稳定遗传。 The leaf-specific senescence-inhibitor gene (PSAG12-ipt) was transferred into Zhong A (an elite indica cytoplasmic male sterile line) and Minghui 63 and Nansheng No.10 (two elite indica restorer lines) by Agrobacterium. It is demonstrated that the foreign PSAG12-ipt gene had been inserted into rice genome and expressed in the resistant HygB rice plants by GUS biochemical analysis, PCR and Southern blot analysis. The antisenescenee results indicate that leaves of some transgenic plants can delay senescence and panicles per plant and weight of 1000-grain are high distinctly compared to the controls. The foreign gene is expressed and heritable in the R1 plants through the analysis of the PCR and PCR Southern.
