摘 要: 4个不同渗透压水平(280、320、360、400 mOsm)的鸡精冷冻稀释液,以甘油(GL)、二甲基亚砜(DMSO)和N,N二甲基乙酰胺(DMA)分别作为抗冻剂进行了冷冻试验.结果表明,添加8%甘油、4%DMSO和11%DMA的冷冻稀释液,4个不同渗透压间冻后精子活率均有显著差异(P<0.05),而且精子活率随冷冻稀释液渗透压的升高而升高;添加5%和10%甘油的冷冻稀释液,渗透压对解冻精子活率影响不显著;在400 mOsm渗透压条件下,8%甘油组冷冻后精子活率高于其他抗冻剂组;3种抗冻剂4个渗透压水平冷冻,平衡时间长短(30 min和90 min)对解冻后精子活率没有影响. Glycerol,DMSO and DMA were supplied in diluents with four different osmotic pressures (280,320,360,400 mOsms)as cryoprotector to frozen chicken's semen. The results showed that. there were significant difference of frozen-thawed motility in each osmotic pressure suplied with 8%GL,4%DMSO and 11%DMA in diluents (P〈0. 05) ,and the motility was promoted with the rise of osmotic pressure; while in diluents with 5 % or 10% glycerol had no significant difference. In this study,the diluents of 400 mOsm gained the highest sperm frozen-thaw motility in three cryoprotector and three glycerol proportion. Among three cryoprotectors only 8 % GL achieved high motility. Under four different osmotic pressures, there was no significant difference between 30 mins and 90 mins to motility(P〉0.05).
