巯基丁二酸镍(Ⅱ)自组装单分子层修饰金电极的制备及H_2O_2传感器的研制

摘  要: 制备了巯基丁二酸镍(Ⅱ)(NiL-SH)自组装单分子层修饰金电极,制做了测定H_2O_2的电流型生物传感器。NiL-SH在电极表面有一个不可逆的还原峰,传感器对H_2O_2的还原具有快速电催化响应(<10s)。采用电子扫描电镜(SEM)对电极表面进行了表征,探讨了电极的电化学反应性质。研究了各种因素,如pH、工作电位等,对传感器响应电流的影响。用循环伏安法测定H_2O_2,线性范围为5.0×10^(-6)~3.0×10^(-3)mol/L(R=0.999),检出限为2.0×10^(-6)mol/L。该电极具有模拟酶特性。米氏常数K_M^(npp)=2.78mmol/L。实验表明,传感器的稳定性、灵敏度和选择性都比较好,可以用于现场测定。 Mercapto-butanedioic add combined nickel (Ⅱ) (NiL-SH) sell-assembled monotayer modified Au electrode was prepared for fabrication of amperometric hydrogen peroxide (H2O2) biosensor. NiL-SH was used as electron mediator and it exhibited an irreversible reduction peak on Au electrode surface. The biosensor has a fast electrically catalytic response to H2O2 (〈10s). The surface of the electrode was characterized by scanning electron microscope and its electrochemical reaction properties were studied. Effects of experimental conditions, such as pH and potential, on the response current of the biosensor were investigated. The linear range is 5.0)K10^-6~8.0×10^-3mol/L (R=0. 999) with the detection limit of 2.0 )〈 10^-6 mol/L for cyclic voltammetry. The electrode showed properties of simulated enzyme. The Michaelis-Menten constant was found to be KM^upp =2.78 mmol/L. The stability, sensitivity and selectivity of the biosensor were investigated. The method has been used to the determination of He O2 in practical samples.
