摘 要: 本实验用β-葡萄糖醛酸苷酶酶解血清样品,采用电喷雾离子源,负离子MRM扫描,建立了血清中司可异罗叶松甘油二酯(SDG)、司可异罗叶松酯素(Seeo)、肠内二醇(EL)、肠内脂(ED)的LC/MS/MS分析方法。本方法线性相关系数r〉0.9991,最低检出浓度(LOQ,S/N=10)分别为SDG50ng/mL,Seco5.0ng/mL,EL5.0ng/mL,ED5.0ng/mL。空白血清加标回收率79.0%~105.6%,变异系数(CV,n=6)〈19.4%。该法灵敏可靠,样品处理简单,适用于血清中SDG、Seco、EL、ED的检测。 A method for determination of Secoisolariciresonol diglucoside(SDG), secoisolariciresinol(Seco), enterolactone(EL) and enterodiol(ED) in serum by LC/MS/MS was established. In this method, fl-Glucuronidase was used to hydrolyze the serum samples, the electrospray ionization sourse and negative ion MRM scan were employed. The linearity correlation coefficients (r) was more than 0.999 1. The limit of quantification (LOQ, S/N = 10) was 50 ng/mL for SDG, 5.0 ng/mL for Seco, 5.0 ng/mL for EL and 5.0 ng/mL for ED. The spiked recovery in blank serum sample ranged from 79.0 % to 105.6 % with coefficient of variation (CV, n=6) less than 19.4%. The method is sensitive and reliable, and the samples handling is simple, which is suitable for determination of SDG,Seco,EL,ED in serum sample.
