摘 要: 目的:肿瘤坏死因子相关凋亡诱导配体(TNF-relatedApoptosis-inducedLigand,TRAIL)受体微阵列生物传感器检测系统为离子敏场效应型(Ionsensitivefieldeffecttransistor,ISFET)免疫生物传感器,利用此检测系统检测细胞表面的TRAIL受体。方法:将1∶10稀释的4种TRAIL受体的抗体吸附在此型免疫生物传感器上,对正常血管内皮细胞株EVC9611、肺癌细胞株AS49-DDP、舌癌细胞株Tca8113以及白血病人来源的白细胞进行检测。结果:这种微阵列传感器检测系统可以同时检测细胞表面的4种TRAIL受体,并发现正常血管内皮细胞株EVC9611和舌癌细胞株Tca8113的细胞表面没有4种TRAIL受体的表达,而肺癌细胞株AS49-DDP细胞表面的TRAILR2表达较多,白血病人来源的白细胞4种受体都有表达。结论:研制的TRAIL受体微阵列生物传感器检测系统可以有效地同时检测各种细胞表面的4种TRAIL受体,为进一步使用TRAIL治疗肿瘤提供基础资料。 Objective:To use microarray bioseusor detection system of TNF-related Apoptosis-induced Ligand(TRAIL) receptors, which is a kind of ion sensitive field effect transistor(ISFET) biosensor, to detect the four kinds of TRAIL receptors on cell surface. Methods:l1 10 diluted four kinds antibodies of TRAIL receptors were, absorbed on the surface of the biosensors. The prepared biosensors with antibody sensitive membrane were used to detect the TRAIL receptors on the surface of human endothelium cell line EVC9611, human lung cancer cell line AS49-DDP, human tongue cancer cell line Tca8113 and the white blood cells(WBC) from a patient with leukemia. Results:This kind of microarray biosensor detection system could be used to detect the four kinds of TRAIL receptors at the same time. The detection showed us that there was no four kinds of TRAIL receptors on the surface of EVC9611 and Tca8113 ; there was TRAIL R2 on the surface of AS49-DDP; there were four kinds of TRAIL receptors on WBC from the leukemia patient. Conclusion: The microarray bioseusor detection system of TRAIL receptors can be used to detect TRAIL receptors on the surface of every kind of ceils, and these data are useful for TRAIL treatment to cancer.
