摘 要: 在乙腈浓度为5%、10%中,木瓜蛋白酶活性比在磷酸缓冲液中显著提高;在乙腈浓度为15%中,木瓜蛋白酶活性与磷酸缓冲液中的基本相等;随着乙腈浓度进一步升高,木瓜蛋白酶活性不断降低。在200 ̄240nm吸收峰区间内,随着乙腈浓度的逐渐升高,木瓜蛋白酶的紫外光吸收值逐渐降低,而且吸收峰也逐渐向长波长方向移动;在240 ̄300nm吸收峰区间内,乙腈溶液中木瓜蛋白酶的紫外光吸收值比在磷酸缓冲液中的高。在200 ̄225nm负吸收峰中,随着乙腈浓度的升高,木瓜蛋白酶的紫外光差示吸收值逐渐降低。乙腈浓度为5%时,木瓜蛋白酶荧光发射峰的荧光强度低于在磷酸缓冲液中的;随着乙腈浓度进一步升高,木瓜蛋白酶荧光发射峰的荧光强度都高于在磷酸缓冲液中的,并逐渐升高,而且发射峰逐渐向短波长方向移动。在5%、10%、20%乙腈中,木瓜蛋白酶的Km、Vmax均高于0%乙腈中的。 The activity of papain was increased remarkably in 5 % and 10 % acetonitrile solution than that in phosphate buffer solution. h, 15 % acetonitrile solution,the activity of papain was equal to that of phosphate buffer solution. With the acetonitrile concentration increasing,the activity of papain reduced constantly. In the absorption peak section of 200-240 nm,the ultraviolet absorption declined constantly with the acetonitrile conccntration increasing,and the absorption peak shifting to long wave. In the absorption peak section of 240-300 nm,tbe ultraviolet absorption in acetonitrile solution was higher than that in phosphate buffer solution. In the negative absorption peak section of 200-225 nm, with the acetonitrile concentration increasing, the differential ultraviolet absorption spectra of papain declined constantly. The value of fluorscence emission peak of papain in 5 % acetonitrile solution was lower than that in the solution without acetonitrile, and the fluorscence emission peak shifting to short wave. With the increasing of acetonitrile concentration, the value increased constantly. In 5 %, 10 % and 20 %(V/V) acetonitrile solution,the Kin, Vmax value of papain were higher than that in 0 % acetonitrile solution.
