摘 要: 以木醋杆菌(Acetobacter xylinum)为出发菌株,细菌纤维素产量为指标,通过正交试验优化发酵培养基,其最优组成为葡萄糖15g/L、蛋白胨10 g/L、酵母膏5g/L、玉米浆40mL/L、磷酸三钠3g/L、柠檬酸钠1g/L、硫酸镁2g/L、氯化钙0.2g/L、硫酸亚铁0.03g/L、乙醇10mL/L。适宜的发酵方式为先130r/min振荡培养10h,后静置培养10d。经验证试验,细菌纤维素产量可达3.81 g/L。 Using Acetobacter xylinum as primary strain, and bacterial cellulose yield as the index, the optimum component proportion was achieved by orthogonal experiment as: glucose 15 g/L, peptone 10 g/L, yeast extract 5 g/L, cream style corn 40 ml/L, Na3PO4 3 g/L, natrium of citrate 2 g/L, MgSO4 2 g/L, CaC12 0.2 g/L, FeSO4 0.03 g/L, and ethanol 10 mL/L. The optimum fermentation condition was: first shaking culture for 10 hours, and then static cultivation for 10 days. The yield of bacterial cellulose could reach 3.81 g/L.
