慢性疲劳综合症差异表达基因筛选的运动实验研究

摘  要: 为了寻找慢性疲劳综合症的标志物,并研究慢性疲劳综合症与身体锻炼之间的关系,对患有慢性疲劳综合症女性的运动前后外周血单核细胞基因表达谱进行了研究。两组女性自愿参加本次实验(即患有慢性疲劳综合症的女性组,称为实验组;身体健康而很少参与体育锻炼的白领组,称为对照组),两组在功率自行车上进行运动,强度控制在其最大负荷的70%左右。在负荷前后抽取血液,分离单核细胞并提取总RNA,逆转录成cDNA,进行信号标记后与带有3 800个寡核苷酸片断的芯片进行杂交。结果是:在负荷前后组内、组间的基因表达都有差异;又用基因功能分类方法对差异进行了评估,实验组与对照组间的运动敏感基因表达的差异主要体现在与染色质合成、核小体、囊泡、细胞骨架以及膜蛋白受体功能有关的基因上;与离子转运和离子通道活性相关基因表达方面,两组间的基准值在运动前就有明显差异,而在运动过后其差异变得更大。 In order to find the signs of chronic fatigue syndrome and study the relation of CFS and exercises, we researched the coenocyte' s gene expression in peripheral blood of women with chronic fatigue syndrome before and after exercises load. Two groups of women took part in this experiment(one was the group of women with CFS, which was called experiment/CFS group, the other was a group of women who were healthy and lack of exercises which was called controlled group). They all have exercises on power bike and the intensity was controlled on about 70% of the maximum load. We extracted their blood before and after their exercises, then separated the monocytes and extracted totle RNA, which were reversely transcribed to cDNAs, and were signed by Cy3 and Cy5 and hybridized with gene chip including 3 800 oligonucleotide snippets. Results displayed: Exercise - responsive genes differed between CFS group and controlled group. We judged differences in gene expression among and between two groups before and after exercises and evaluated differences in terms of Gene ontology comparison. These were in genes classified in chromatin and nucleosome assembly, cytoplasmic vesicles, membrane transport, and G protein- coupled receptor ontologies. Differences in ion transport and ion channel activity were evident at baseline and were exaggerated after exercise, as evidenced by greater numbers of differentially expressed genes in these molecular functions.
