摘 要: 利用高盐低pH值法对杨树叶绿体及其DNA进行分离与提取,经匀浆—差速离心—裂解—纯化,可获得完整的叶绿体和纯度较高的叶绿体DNA(chloroplastDNA,cpDNA),DNA平均产率0.64ugg/.本方法快速简便、省去传统的蔗糖密度梯度离心和氯化铯密度梯度离心等昂贵、耗时的步骤,对仪器的要求不高,成本低,适用于实验教学和科研活动. The method is improved by the use of high salt in combination with low pH buffer to isolate chloroplast and DNA from leaves of populus. The process include whipping, differential centrifugation, cracking and purification. It can be obtained that high quality chloroplast and higher yield 0. 64 ug/g cpDNA. The technique is easy to obtain cpDNA with high yields, high purity and high quality, it avoids the timeconsuming and expensive process of Cesium Chloridesucrose density gradient centrifugation or sucrose step gradients centrifugation.
