摘 要: 辅酶Q10是一种重要的类维生素有机物,是促进生物氧化的酶类生化药物。通过L16(45)正交实验,研究皂化法提取辅酶Q10的工艺条件;用硅胶柱层析及制备硅胶板分离纯化;用薄层色谱、紫外光谱对提纯样品进行定性及定量分析。结果表明:皂化法提取罗非鱼肝脏辅酶Q10的最佳工艺条件是:皂化温度70℃、皂化时间30min、萃取4次。经硅胶柱层析,每100g罗非鱼肝脏可以提取纯化辅酶Q103.39mg。 The coenzyme Q10 (Co Q10) is a kind of fat soluble organic matter has some important biological activity. It is biochemical medicine of bio-oxidase. A method of extracting and purification of Co Q10 from Tilapia liver are developed. The technology conditions of extracting Co Q10 from Tilapia liver through saponification by orthogonal test L16 (4^5), the method of purity Co Q10 through Silica gel chromatogramphy and Silica gel thin layer chromatography, and determined by TLC, UV were studied. The results showed that the optimal extracting conditions of Co Q10 from tilapia liver are: wet culture distilled at 70 ℃ for 30 min, and extracted with petroleum ether four times. 3.39 mg purified Co Q10 was gained from 100 g Tilapia liver by Silica gel chromatogramphy. An accurate analysis result was obtained by using TLC separation and UV absorbance.
