摘 要: 选择具有高保真性的Taq plusⅠ DNA聚合酶,以小鼠表皮生长因子mRNA为模板研究其反转录性能。对cD-NA的检验结果表明,Taq plusⅠ DNA聚合酶在72℃下有良好的反转录活性,且由于其高保真性,故在进行反转录时既可避免碱基错配,又可避免在低温下用AMV反转录酶或Mulv反转录酶进行反转录时可能造成的cDNA序列缺失。 In this research the Taq PlusⅠDNA polymerasc with satisfying fidelity was selectred to study its reverse transcription activity using mouse epidermal growth factor mRNA as template.The results of cDNA product showed that the enzyme has a satisfying reverse transcription activity in 72C,and with its satisfying fidelity,it is concluded that using Taq plusⅠDNA polymerase could avoid not only the wrong complement of base but also the probable sequence deletion of cDNA if AMV or Molv reverse transcription enzyme used.
