紫果西番莲的离体形态发生
Morphogenesis of Passiflora edulis in Vitro

摘  要: 以紫果西番莲的腋芽萌发的芽苗为基础试材,以顶芽、茎节间和叶柄作为外植体,培养在改良MS培养基(大量元素减半)附加IBA1—4mgL-1、蔗糖2%的培养基上诱导生根,在IBA2和4mgL-1的条件下,顶芽生根率均为100%,在IBA2mgL-1的条件下,茎段和叶柄的生根率达80%;茎节间、根段和叶柄作为外植体,培养在改良MS培养基(大量元素减半)附加ZEA2、2.5或3mgL-1,蔗糖2%的培养基上诱导不定芽,在ZEA2,2.5和3mgL-1的条件下,茎段的长芽率均为100%,在ZEA2.5mgL-1的条件下,根段和叶柄的长芽率分别为33%和37% Shoots of Passiflora edulis germinated from axillary buds were used as experimental material for morphogenesis in vitro. Terminal bud, internode segment and leaf stalk of P. edulis, all of which were cut from the shoots, were cultured on modified MS medium (macroelements being half strength) supplemented with 2% sucrose and 1-4 mg·L -1 IBA. The most effective rooting treatment was achieved with terminal buds cultured in 2 or 4 mg·L -1 IBA, the induction rate of rooting being as high as 100%, and rooting occurred in as early as 10-12 d; while the rooting rate both internode segment and leaf stalk cultured in 2 mg·L -1 IBA were 80%. Internode segment, root segment and leaf stalk explants were cultured in modified MS basal medium (macroelements being half strength) added with 2% sucrose and 2-3 mg·L -1 ZEA, the most effective budding treatment was achieved with internode segment, the induction rates of the shoots was 100% in either 2, 2.5 or 3 mg/L ZEA,33% and 37% of induction rate of the shoots were attained with root segment and leaf stalk in 2.5 mg·L -1 ZEA, respectively, higher than that in 2 or 3 mg·L -1 ZEA.
