摘 要: 参照Kajiwara等人获得的β-胡萝卜素酮化酶cDNA序列设计引物,利用RT-PCR技术扩增雨生红球藻的β-胡萝卜素酮化酶cDNA的完整编码片段并进行了序列测定与分析.结果表明,所获得cDNA序列与Kaji-wara等人报道序列具有96.4%的相似性,蛋白序列具有98.3%的相似性,说明所获得的cDNA序列确为β-胡萝卜素酮化酶的cDNA序列. A pair of specific primers of β-carotenoid ketolase(crtW) had designed according to the cDNA sequence reported by Kajiwara, and then its cDNA complete reading frame had been cloned from Haematococcus pluvialis by RT-PCR and sequenced. Sequence alignment shows that the similarity between the cDNA sequence obtained and sequence reported by Kajiwara is 96.4 % and the'protein similarity is 98.3 %, which suggests that the cDNA obtained should be the crtW coding sequence.
