参麦注射液对体外培养软骨细胞增殖及DNA合成的影响

摘  要: 目的:探讨参麦液对软骨细胞增殖与DNA合成的影响。方法:运用关节软骨体外培养方法,设立空白对照与IGF-Ⅰ药物对照组,采用M TT比色法与二苯胺显色法观察不同浓度参麦液对软骨细胞增殖与DNA合成情况。结果:参麦液与生长因子IGF-Ⅰ均可促进软骨细胞增殖与DNA合成,同空白对照组相比具有非常显著性差异(P<0.01),其中5%参麦液组同浓度为5 ng/m l的IGF-Ⅰ组作用效果相当(P>0.05)。结论:参麦液可促进软骨细胞的合成代谢,减少其分解代谢,从而减轻关节软骨的退变,延缓OA的进展。 Objective:To study the effect of shenmai liquid on the proliferation and DNA synthesis of chondrocyte. Methods:Joint chondroctye was cultured in vintro and randomly distributed into eight groups, that is the blank control group, the 1 percent shenmai liquid group, the 2.5 percent, the 5 percent, the 7.5 percent, the 10 percent, the IGF- Ⅰgroup(5ng/ml) and the IGF- ⅠⅡ group (10ng/ml) . MTT and diphenylamine method were adopted in the study. Results: Both shenmai liquid and IGF- Ⅰcoulcl facilitate the proliferation and DNA synthesis of chondrocyte. There was great significant difference compared with the blank group(P〈0. 1). There was no difference between the two groups of 5 percent shenmai liquid and IGF- Ⅰ group (5ng/ml)(P〉0.05). Conclusions: Shenmai liquid could both promote anabolism and decrease catabolism of the chondrocyte , thus it could defer the degeneration of osteoarthritis cartilage.
