摘 要: 为利用蛋白纯化技术获得较高纯度美洲大蠊天然主要变应原,以进一步提高临床诊断的特异性和免疫治疗疗效。美洲大蠊全虫粗浸液通过DE-52离子交换层析、HiprepSephacrylS-200凝胶过滤层析等分离步骤得到了纯度为90%的目的蛋白,回收率为26%,经WesternBlotting分析74kDa的蛋白质具有免疫原性,为变应原疫苗的纯化和标准化奠定了基础。 To isolate and identify the major allergen of Periplaneta americana, Sequential purification of two steps including DEAE-cellulose 52 ion exchange chromatography and Hiprep Sephacryl S-200 gel filtration chromatography were carried out to obtain the purified major allergen from the crude extract of Periplaneta americana. Western Blotting analysis showed the obtained protein with 74kDa was the major allergen of Periplaneta americana, the harvesty of purity target allergen were up to 90%. So the procedure should be a improved method to obtain purified allergen from Periplaneta americana.
