摘 要: 建立了高效液相色谱(HPLC)测定鸡肝中洛克沙砷含量的实验方法.样品经匀浆、甲醇提取、高氯酸去蛋白后,用Nova-Pak色谱柱(C18,150×3.9mm,4μm)分离,以0.05 mol/L磷酸二氢钾:甲醇:10%乙酸(80:10:10)为流动相,在流速0.8 mL/min,柱温30℃,进样量10μL的条件下,于264 nm波长处检测.结果表明,该方法的线性范围为0.50~10.0μg/mL,线性相关系数R2=0.9741,在两个添加浓度水平上的平均加标回收率分别为83.7%、89.9%. A high performance liquid chromatographic method (HPLC) was developed for the determination of roxarsone in chicken livers. The liver samples were subjected to homogenate and extraction by using methanol as extracting agent, and then HClO4 was utilized to get rid of protein. The chromatographic separation was performed on a Nova-Pak C18 column ( 150 × 3.9 mm,4 μm) using 0.05 mol/L KH2PO4: CH3OH: 10% CH3COOH(80: 10: 10) as mobile phase at a rate of 0.8 mL/min, temperature of 30℃, injection sample size of 10 μL and UV detection at 264 nm. The results demonstrated that the linear range was 0.50 - 10.0 μg/mL with linear relative coefficient 0. 9741. The average recoveries of two different addition levels were 83.7% and 89.9% respectively.
