摘 要: 采用一种灵敏、简单的方法, 可实现对β-人绒毛膜促性腺激素(β-HCG)的高通量检测.本法具有酶联免疫吸附(ELISA)法的特异性、增强化学发光法(ECL)的高灵敏度及化学发光成像分析高样品通量等优点.冷却电感耦合 (CCD)成像系统被用于检测增强化学发光信号.发光强度值与β-HCG在12.5 ~ 400 mIU/mL范围内呈线性关系;检出限为4 mIU/mL.测定了妇女尿液中β-HCG的含量,分析结果与医院的诊断结果一致. A sensitive and simple method has been used for high throughput detection of β-HCG (β-human chorionic gonadotropin). A typical sandwich type imunoassay was used. The proposed method has the advantage of showing the specificity of enzyme-linked imrnunosorbent assays (ELISA), sensitivity of enhanced chemiluminescence (ECL) , and high throughput of chemiluminescence (CL) imaging. A commercial highsensitivity cooled charge coupled device (CCD) camera has been applied to image the weak chemiluminescence from the ECL. The linear range of β-HCG was 12.5 to 400 IU/L (r =0. 9983) and the detection limit was 4 IU/L. β-HCG contents in women urine were determined and the analysis results agreed well with those from hospital.
