摘 要: 采用"直接水提法"以及"KCl分级法"来提取琼枝和异枝麒麟菜中的多糖.分别用蒽酮-硫酸法、间苯二酚法、明胶-BaCl2分光光度法测定多糖样品的总糖质量分数、3,6-内醚半乳糖质量分数、总硫酸基质量分数;用凝胶色谱法测定各多糖样品的相对分子质量分布情况;用GC-MS测定麒麟菜硫酸酯基多糖的单糖种类及含量.结果显示采用"直接水提法"以及"KCl分级法"比传统的"碱处理法"产率高,而且硫酸基质量分数也明显增高.两种麒麟菜多糖均为硫酸酯基多糖,大部分都属于κ-卡拉胶类.各多糖样品的相对分子质量分布在105左右.麒麟菜多糖主要是由半乳糖和3,6-内醚半乳糖组成,此外还含有少量的葡萄糖、木糖、塔罗糖和艾杜糖等,不同方法提取的多糖样品其单糖种类基本一致. The water extraction method and the KCl classification method was used to extract the sulfated polysaccharides. The anthrone- sulfuric acid method, the resorcinol method and the gel-BaCl2 spectrophotometry method were separately used to determine the total sugar content, the 3,6 - AG content and the content of the SO4^2- . Furthermore, the GC-MS was used to analyze the kinds and the content of the monosaccharide. The results indicated that the yield rate of the water extraction method and KCl classification method are higher than the base solution method, moreover the SO4^2- content is higher also. Both the polysacchafides are sulfated polysaccharide, most of them are κ-carrageenan. The molecular weights of the polysaccharides are about ten thousand. The polysaccharide are mostly composed of 3,6 - AG and a few glucose, xylose, talose and idurose. The polysaccharides extracted by different method have similar monosaccharide content.
