摘 要: 以采自汕头的南美白对虾为研究对象,采用亲和层析、PAGE、SDS-PAGE、Western-blotting、血细胞凝集实验和糖抑制实验等方法探索南美白对虾血蓝蛋白的血细胞凝集活性.结果发现,血蓝蛋白对实验所选用的鱼、鸡、鼠和人等4种红细胞均具有显著的血细胞凝集活性,凝集活性大小为5~10mg/L,而且其凝集反应可被α-半乳糖、α-D葡萄糖、甘露醇和N-乙酰神经氨酸等4种糖所显著抑制.由此推测,血蓝蛋白确实具有血细胞凝集活性,这对进一步研究血蓝蛋白的免疫学功能具有重要意义. An attempt was made to testify whether hemocyanin purified by affinity chromatography possesses agglutinative function with erythrocyte,The methods of PAGE,SDS-PAGE, Westernblotting, agglutination reaction and carbohydrate inhibition agglutination test were used, in which four types of erythrocyte including fish, chicken, mouse, human and nine kinds of carbohydrate including α-D-Lactose, Sucrose, α-D-Glucose, Maltose, D-Galactese, D-Xylese, D-Sorbese, Mannitol, N-Acetylneuraminic acid were selected to analyze respectively. The results showed that hemocyanin has aggregation with all the four types of erythrocyte, whose agglutinative activity was about from 5 to 10 mg/L. Furthermore, the agglutinative activity of hemocyanin could be inhibited by α- Lactose, α-D-Glucose, Mannitol and N-Acetylneuraminic acid respectively. These results suggested that hemocyanin indeed has agglutinative function, which will be helpful for the research of hemocyanin' s immue functions.
