壳聚糖-激素药条埋植技术初探
Preliminary studies on the chitosan-hormone implantation

摘  要: 采用壳聚糖-激素共沉淀方法,制备了新型的壳聚糖-甲基睾酮(chitosan-MT)激素埋植药条.药条埋植于实验鱼8天和15天后分别取血样检测GtH含量和解剖实验鱼观察药条的缓释情况,采用放射免疫检法检测血清GtH含量,结果表明,chitosan-MT埋植8天和15天后雄鱼的血清GtH平均含量分别提高到原来的4.36倍和4.53倍;而雌鱼的GtH含量分别是实验前的2.76倍和2.74倍;但同时期取样的雌、雄对照组鲤鱼血清GtH平均含量没有明显差异.由此可见,制备的chitosan-MT药条能在鲤鱼机体内稳定而缓慢释放,并能通过正反馈途径促进GnRH的释放,从而使血清GtH含量增加. Novel chitosan-methyl testosterone (chitosan-MT) bars were prepared by co-precipitation method. The controlled release of methyl testosterone in chitosan-MT bars was investigated and GtH concentration was assayed by using the radioimmunoassay (RIA) after they were implanted into the tested carps. The results showed that the mean GtH concentration in the male carps serum was increased by 4.36 and 4.37 times after 8d and 15d implantation of chitosan-MT bars, while the mean GtH concentration in the female carps serum was 2.76 and 2.72 times of the origin after the same treatment. However, the mean GtH concentration of serum remained relatively constant in the control carps. So it was deduced that MT was able to be released slowly in the chitosan-MT bars implanted in the carps and then promoted gonadotropin-releasing hormone (GnRH) release by a positive feedback adjustment, which finally indirectly resulted in an increase of the GtH content in serum.
