摘 要: 磺胺二甲嘧啶(SM2)7个剂量(30~400mg/kg),新西兰白兔25只,共63次快速静注,测定SM2在兔体内的动力学是否存在着非线性的特征。血中SM2及其N4-乙酸化代谢物(N4SM2)用重氮化-偶合比色法测定,药-时数据用药物动力学和统计学微机程序处理。结果表明:SM2在兔体内的分布和乙酸化代谢、N4SM2从兔体内的消除均存在着某种非线性特性;用线性动力学模型处理具有非线性动力学因素的药-时数据掩盖药物的非线性的特征。这些结果对兽医药物动力学研究和临床上合理用药有参考意义。 Sixty three pharmacokinetic experiments of sulfadimidine (SM2) intra-venous injection, with seven dose levels (30~400 mg/kg) of SM2, were conducted on twen-ty five New Zealand white rabbits in order to determine whether or not there exist nonlinear features of SM2s pharmacokinetics in rabbits. Concentrations of SM2 and its N4-acetyl metabolite (N2SM2) in plasma were analysed with spectrophotometry. The concentration-time date were processed with pharmacokinetic and statistical micro-computer programs. Re-sults showed that there were some phenomena of the distribution and acetylation of SM2 in rabbits, and of elimination of N4SM2 from the body. Marked differences of pharrnacokinetic parameters of SM, between different rabbits were observed with dose changes. It is impossi-ble to take ture nonlinear properties of a drug when one uses a linear pharmacokinetic model to process the concentration-time data of the drug whose disposition includes some nonlinear kinetic factors. The results have significances for veterinary pharmacokinetic studies and clinically reasonal administration of drugs.
