摘 要: 以氧合血红蛋白为原料,分别用酸性丙酮萃取—减压蒸馏和酸性丙酮萃取—NaAc沉淀两种方法制备氯化血红素。产品用吡啶—NaCl的醋酸饱和溶液重结晶进行纯化,通过元素分析,红外光谱和快原子轰击质谱对血红素进行确证。发现粗品中存在着与血红素铁配位的小分子(离子或基团)杂质。这些杂质可以通过重结晶的方法除去。最后还对血红素的质谱裂解机理做了探讨。 Heme is an important kind of natural porphyrin.Thus,it is very sinificant to modify hemeand investigate catalytic and pharmacological activities of the derivatives thus obtained。In thiswork,hemin was prepared fromhemoglobin by two different methods,purified byrecrystallization in pyridin-Nacl- CH_3COOH solution,and identified by element analysisIR-spectrum and fast bombardment mass spectrum(FAB-MS).Agood hemin yelid withhigh purity could be obtained in this preparation and the impurities bonded to theporphyrinoiron were removed by recrystailization。An explanation for the fragment ions of thepositive ion spectrum was given with reference to FAB splitting rules of organic compounds。
