摘 要: 以电场强度12.5kv/cm,RC设定值为5ms为条件,将pBC11质粒DNA从枯草杆菌向大肠杆菌电击转移,一次可得220个转移子。大肠杆菌种内的质粒电击转移,一次可得4200个转移子。但质粒从大肠杆菌向枯草杆菌以及枯草杆菌种内的质粒电击转移效率都较低。实验表明,高电场脉冲可使质粒DNA突破细胞间的屏障实现直接转移。 Plasmid pBC11 was transferred directly form B.subtilis to E.coli by high voltage electroporation.At a single pulse of 12.5KV/cm,5msec.220 transfer colonies were obtained perelectroporation,4200 colonies in intraspecific transfer of E.coli. In intraspecific transfer of B.subtilis and interspecific transfer from E. coli to B.subtilis transfer efficiencies were lower.Re-sults showed that Electrotransfer has the potential to transfer plasmid DNA across bacterialspecies barrierrs.
