“渝苏303”甘薯离体形态发生过程中生理生化特性的变化
Changes in Physiological and Biochemical Characters of Sweet Potato Cultivar Yusu 303 during in vitro Morphogenesis

摘  要: 甘薯茎段、叶片和叶柄在附加有1.0mgLNAA的MS培养基中培养5d可产生大量的不定根,培养20d茎段和叶片分化出不定芽。本文以甘薯品种“渝苏303”为材料,研究了甘薯外植体在不定根和不定芽分化过程中可溶性蛋白质含量、过氧化物酶(POD)和超氧化物酶(SOD)活性的变化,以及可溶性蛋白质和SOD同工酶酶谱的变化。结果表明,在甘薯叶片外植体不定根分化过程中,POD活性呈上升趋势,而可溶性蛋白质含量和SOD活性呈下降趋势。在不定芽形成过程中,可溶性蛋白质含量呈下降趋势,而POD和SOD活性呈上升趋势。可溶性蛋白质和同工酶酶谱分析表明,在甘薯茎段离体形态发生过程中出现了3条新的可溶性蛋白质谱带,且谱带颜色逐渐变深。在不定根形成前,POD同工酶酶带为8条,而在不定根开始形成时及形成以后,出现了3条新的POD同工酶酶带,其Rf值分别为0.50、0.67和0.68。 The development of a simple and efficient way for plant regeneration in vitro is a basis in sweet potato breeding by gene engineering. In order to reveal the relationship between the changes of physiological characteristics and the induction of adventitious roots and buds, sweet potato cultivar Yusu 303 was used to analyze the changes of water-soluble protein content, activities and electrophoresis banding patterns of peroxidase (POD) and superoxidase dismutase (SOD) isoenzymes during adventitious root and bud formation. The results showed that stem, leaf and petiole explants of sweet potato cultured on MS medium with 1.0 mg/L NAA produced great number of adventitious roots in 5th day, and stem and leaf explants produced adventitious buds at 20th day, respectively, during in vitro morphogenesis. The results showed that during adventitious roots formation, activity of POD in leaf explants was increased, but the content of water-soluble protein and the activity of SOD decreased. During bud differentiation, the content of water-soluble protein in leaf explants was decreased, but the activities of POD and SOD increased. The results of electrophoresis banding patterns indicated that there were three new bands of water-soluble protein in stem explants during root and shoot formation, and their color was deepened. There were eight bands of peroxidase isoenzymes in rootless shoots before the adventitious roots and shoots formation, and three new bands of peroxidase isoenzymes occurred in the stage of root and shoot formation, their R_f values were 0.50, 0.67 and 0.68, respectively. The contents of water-soluble protein, activities and electrophoresis banding patterns of peroxidase (POD) and superoxidase dismutase (SOD) isoenzymes may be regarded as physiological indexes of in vitro morphogenesis in sweet potato.
