摘 要: 采用4%N aC l培养基选择分离法,从我国10个烟区采集的801个复烤烟样中,分离出苏云金芽孢杆菌736株。分离出的Bt菌经PC R毒素蛋白基因鉴定,其中含cryⅠ毒素蛋白基因的Bt菌有7株,出菌率为0.95%;含cryⅤ毒素蛋白基因的Bt菌21株,出菌率为2.85%;没有含cryⅢ毒素蛋白基因的Bt菌。对含毒蛋白基因cryⅠ或cryⅤ的28个Bt菌株用二龄烟草甲虫进行生物毒力测定,试验后9d有18个Bt菌株生物毒力测定校正死亡率均超过80%;试验后12d有9个Bt菌株生物毒力测定校正死亡率均超过95%。可见,用苏云金芽孢杆菌防治烟叶仓贮害虫是有潜力的。 Bacillus thuringiensis strains were isolated from 801 tobacco sample collected from 10 tobacco production area in China by the method of 4%NaCl cutural medium.The identification results of PCR toxin gene indicated that 7 Bt isolates with plating efficiency of 0.95% contained cryⅠ toxin gene 21 Bt strains contained cryⅤ gene without Bt isolates containing cryⅢ gene. The bioassay toxicity of 28 Bt isolates containing cryⅠand cryⅤ gene was determinated to Lasioderma serricorneF. second instar. The results showed that the corrected mortality of 18 Bt exceeded 80% 9 days after inoculation and 9 Bt isolates exceeded 95% 12 days after inoculation.
