摘 要: 对人工饲养的北星二号蚯蚓纤溶酶的分离纯化表明,其较佳工艺为磷酸缓冲液保温或匀浆提取,Amberlite IRA—904或DEAE—纤维素柱层析,Sephades G—100凝胶过滤,真空冷冻干燥。按此工艺可得到纤溶化活力为83700mm^2/mg,蛋白的蚯蚓纤溶酶。 Artificially fed earthworms (Eisenia foetida) were used as experimental material for separating and purifying earthworm fibrinolysin. A comparatively satisfactory technology was developed through screening and comparing Id kinds of ion exchange resins and several kinds of DEAE -cellulose. Its main steps are as follows; pounding the fresh earthworm into small pieces, homogenizing them before extraction of incubating them with phosphate buffer solution, separating by ion exchange chromatography with Amberlite IRA - 904 or DEAE - cellulose, filting with Sephadex G- 100 gel and vacuum freeze -drying. Requiring only simple equipment and easy and convenient to manipulate, this technology is suitable for large - scale production of earthworm fibrinolysin.
