The purified lectin from Phaseolus coccineus var.rubronanus(PCI) showed a major protein band and a thin, minor one with less mobility by PAGE,but only one band by SDS-PAGE with or without meroaptoethanol. This thin band disappeared when Gly-HCI buffer containing mannan was used for eluting PCL from the thyro globulin-Sepharose affiniry column.However) the use of mannose or arabinogalaotan did not affect this thin band.Also the PAGE pattern of PCL when further purified on gel filtration or ion exchange chromatographie column was not changed. Both the H-oligosaeeharides(H-Man8 XylGlcNAc2(Ⅰ) and 3H-Man6Xy1Gl1cNAc2Fuc(Ⅱ)) prepared from PCL bound to immobilized PCL column and could be eluded from the column by phosphate buder containing mannan,thus demontwating the formation of intermoleeular polymerization through non-covalent binding of the oligosaccharide of one protein molecule to the carbohydratesbinding site of another molecule of lectin.
