摘　　要： 本文提出了一种偶合反应化学发光酶联免疫分析测定人血清甲胎蛋白的新方法。该法将辣根过氧化物酶(HRP)催化H_2O_2氧化曙红(Eosin)的反应和HRP催化H_2O_2与发光剂鲁米诺(Luminol)的化学发光反应相偶合,根据测定标记在甲胎蛋白抗体上的HRP的量,求得发生免疫反应的甲胎蛋白抗原的含量。测定范围为0.10～100.0ng/ml甲胎蛋白,比现行的ELISA法灵敏度高10倍,且两法的相关性很好。 A new chemiluminescence enzyme immunoassay with coupling reaction for determining the a-fetoprotein (αFP) in serum is developed. It is based on coupling reaction of eosin-H_2O_2 catalyzed by horseradish peroxidase @-fetoprotein anti-body (HRP-αFPAb) with the chemiluminescence reaction of Iuminol-H_2O_2 catalyzed by HRP-αFPAb to measure the activity of HRP-αFPAb. The double antibody sandwich immunological method is used for the determination of αFP. The linear relationship of the method holds for αFP within 0.1 -100. 0 ng/ml. The current enzyme-linked immunosorbent assay(ELISA) absorptiometric detection is taken as a comparison method and similar results have been obtained (r=0. 998). The sensitivity of the method is improved 10 folds comparing with current ELISA method.
