摘 要: 以盐藻总RNA为模板,根据已获得的盐藻PSY基因起始和终止密码子附近的核苷酸序列设计引物,通过RT-PCR扩增获得盐藻PSYcDNA片段.盐藻PSY基因cDNA全长1260bp,编码的盐藻PSY蛋白由420个氨基酸组成,大小为47.3kD,等电点为8.67.在GenBank中进行BlastP检索,发现该片段与许多植物PSY具有较高的相似性(78%~89%).系统进化分析进一步证明,藻类PSY与蓝细菌PSY亲缘关系最近. Amplification by RT PCR using D.salina RNA as temple was done with specific primers that were located around the start codon and stop codon of PSY DNA sequence of D.salina. The obtained PSY cDNA fragment of D.salina is 1 260 bp in length. The deduced amino acid sequence consists of 420 amino acids and the polypeptide was 47.3 kD whose pI was 8.67. BlastP search in GenBank showed that PSY of D.salina shared high similarity with that in plants up to 78%~89%. Phylogenic analysis showed that PSY of species in Dunaliella had a close relationship with those of Cyanobacterium.
