摘 要: 应用AFLP技术对采自广东稻区7个地点不同生物型稻瘿蚊DNA进行指纹分析,并用DPS软件进行聚类分析,结果表明,AFLP可对稻瘿蚊DNA扩增出丰富的指纹带,不同稻瘿蚊种群和生物型DNA带型存在共同性,也有特异性;不同引物扩增数据用DPS软件分析得出不同的聚类结果。对4组引物的分析结果表明,只有1组引物所扩增的广东7个地点4个稻瘿蚊生物型DNA指纹可以进行较清晰聚类,说明鉴定稻瘿蚊生物型需要找到适合的引物组合,并指出利用DNA指纹技术分析稻瘿蚊生物型时存在一定的不确定性。 In this study, AFLP analysis was used to assess the DNA fingerprints of gall midge biotypes from seven different localities of Guangdong Province, and cluster analysis was performed by DPS software. The results showed that DNA fingerprints of gall midge were plentiful by AFLP analysis, the DNA bands of different gall midge biotypes were different, the cluster results by using different primers were different. Four groups of primer were used to AFLP analysis, only one had distinct cluster result. The result showed that suitable primer was essential to identify gall midge biotypes, so uncertainty was existed when DNA fingerprints was used to analyze gall midge biotypes.
