摘 要: 利用RAPD技术对苦瓜杂种一代顶优苦瓜及其父母本的特异性进行分析,结果引物OPU19扩增的OPU19-1600bp片断在父本和F1中出现,在母本中不出现;对该一代杂种的F2、B1、B2代进行的遗传分析表明:OPU19-1600bp片断可能与父本的某一对基因连锁,在F2、B1、B2代中符合孟德尔一对显性基因的分离规律,从而证实其可用于区分杂交种与母本的自交种。 RAPD (random amplified polymorphic DNA) analysis was employed in order to study the specific fragment in balsam pear (Momordica charantia L.) hybrid F_1'DingYou'and it's parents. The results showed that primer OPU19 amplified one OPU19-1600 bp fragment which appeared in F_1 and male parent, but not in the female parent. The analysis of F_2 and backcrosses progeny revealed that this fragment might be linked with one gene of male parent, and it conform to the Mendelian inheritance of one pair of dominant gene. Thus OPU19-1600 bp could be used to test the seeds of F_1 hybrid and inbred of female parent.
