摘 要: 参考GenBank中发表的猪源O型口蹄疫病毒 (FMDV) 3ABC的基因序列 ,设计一对特异引物 ,分别以 5株猪源O型FMDV流行毒株基因组RNA为模板 ,通过RT_PCR的方法获得 3ABC基因 ,并克隆到pMD18_T载体中。测序结果显示 :5株O型FMDV流行毒株的 3ABC基因cDNA均为 12 81bp ,编码由 4 2 7个氨基酸残基组成的多肽。同源性比较和系统发育树分析表明 :5株O型FMDV流行毒株亲缘关系密切 。 In this study, five FMDV type O epidemic strains were gathered.RT-PCR was employed to amplify the cDNAs of the 3ABC gene of five strains with a pair of specific oligonucleotide primers (3A/3C).The cDNAs were cloned into the T-T windows of plasmid pMD18-T vector respectively.The inserts were sequenced and the results showed that the full-length sequences of the 3ABC gene of all the five strains consisted of 1281 nucleotides (nt) respectively,coding for predicated polypeptides of 427 amino acid residues.Analyses of the 3ABC gene nucleotide sequences and deduced amino acid sequences suggested that five FMDV type O epidemic strains shared (100?%) identity.Comparison of the homologies of the 3ABC genes showed that there was 96.8?%~98.4?% nucleotide sequences homology and 97.2?%~98.8?% deduced amino acid sequence homology among the five strains.Based on the results of nucleotide sequence and deduced amino acid sequence homologies,the phylogenetic trees of the 3ABC gene were constructed.The positions of the five strains were highly related between the two trees.
