大鼠骨髓细胞诱导分化神经细胞的形态学观察

摘  要: 本实验探讨了从骨髓诱导培养分化神经细胞的方法,并从形态学上观察了其演变规律.抽取SD大鼠骨髓做全骨髓细胞培养,用光镜、扫描电镜观察不同阶段的形态变化,用免疫细胞化学方法进行鉴定.观察到骨髓细胞由单细胞、细胞分裂、形成克隆团(球)、出芽,直至形成突起成为神经元样细胞的演变过程.可确定神经元样细胞的前体在骨髓阶段是一类'大圆细胞'.24h左右开始贴壁,前体细胞长出'足'样结构与细胞外基质相连接.培养25~35 d细胞分化为神经元样,神经元样细胞的形态多样,有球形、锥形、星形或梭形等,直径在15.3~36.5μm之间.应用免疫细胞化学鉴定出神经干细胞、神经元和胶质细胞.结果表明,骨髓细胞可分化为神经细胞,扫描电镜从超微结构上给予佐证.为骨髓细胞向神经细胞分化的发育过程提供了形态学资料. The present study was designed to find a method of neural cell differentiation from the bone marrow cells and observe their morphological changes. The bone marrow cells were cultured from aspirates taken from the femur and tibia of albino Sprague-Dawley adult rats (Harlan-Sprague-Dawley), The morphological changes of these cells in different stages were observed by light microscope and scanning electron microscope, and then identified by immunocytochemical staining. The results showed that the developmental process from bone marrow cells into the neuron-like cells included monocellular, cell division, clone mass or sphere formation, budding and apophysis formation, which were observed by scanning electron microscope. These data may infer that a kind of big and round cells (BRC) is possibly the precursor of neuro-like cells in bone marrow. Cells erupt foot-like structure and adhered to the extracellular matrix (ECM) at 24 hours and differentiating neuro-like cells at 20-30 days after culture. Neuro-like cells have various forms, including spherical shape, taper, asteroid and fusiform shape. The neural stem cells, neurons and glial cells were identified as positive for nestin, NeuN and GFAP respectively. The experiment was successed in finding a method of differetiation from the whole bone marrow cells into neural cells, and the ultrastructures of these cells were confirmed by scanning electron microscope. The study provided the morphological data of the differentiation from bone marrow cells into neural cells.
